چکیده

A novel, cell-surface protein essential for Ca(2+) release-activated Ca(2+) (CRAC) channel function has been identified through independent genome-wide screens. This huge advance will enable molecular dissection of the CRAC channel complex, moving the field beyond Icrac signature to structure.

Calcium Signaling: Pyruvate and CRAC Meet at the Crossroads

Receptor-evoked Ca(2+) influx is mediated by store-operated Ca(2+) channels, such as the CRAC channel, which mediates the I(crac) current. Recent work reveals that pyruvate, the precursor substrate for the Krebs cycle, regulates I(crac) to prolong Ca(2+) influx into the cell, thereby coupling oxidation of glucose and its own metabolism in the mitochondria to Ca(2+) influx by the CRAC channel.

Chlamydomonas CAV2 Encodes a Voltage- Dependent Calcium Channel Required for the Flagellar Waveform Conversion

Cilia and flagella can alter their beating patterns through changes in membrane excitation mediated by Ca(2+) influx. The ion channel that generates this Ca(2+) influx and its cellular distribution have not been identified. In this study, we analyzed the Chlamydomonas ppr2 mutant, which is deficient in the production of a flagellar Ca(2+) current and consequently has a defective photophobic res...

STIM1 Is a Calcium Sensor Specialized for Digital Signaling

When cells are activated by calcium-mobilizing agonists at low, physiological concentrations, the resulting calcium signals generally take the form of repetitive regenerative discharges of stored calcium, termed calcium oscillations [1]. These intracellular calcium oscillations have long fascinated biologists as a mode of digitized intracellular signaling. Recent work has highlighted the role o...

Therapeutic Discovery ORAI1-Mediated Calcium Influx in Lactation and in Breast Cancer

The entry of calcium into the mammary epithelial cell from the maternal plasma (i.e., calcium influx mechanisms) during lactation is poorly understood. As alterations in calcium channels and pumps are a key feature of some cancers, including breast cancer, understanding these calcium influx pathways may have significance beyond mammary biology. We show that the store-operated calcium influx pro...

Target-Cell Contact Activates a Highly Selective Capacitative Calcium Entry Pathway in Cytotoxic T Lymphocytes

Calcium influx is critical for T cell activation. Evidence has been presented that T cell receptor-stimulated calcium influx in helper T lymphocytes occurs via channels activated as a consequence of depletion of intracellular calcium stores, a mechanism known as capacitative Ca(2+) entry (CCE). However, two key questions have not been addressed. First, the mechanism of calcium influx in cytotox...
